ADPN Antibody (D-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects ADPN protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-ADPN antibody (D-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ADPN, a member of the Adiponutrin family, plays a crucial role in lipid metabolism due to lipase activity, which depends on an activated serine residue. This enzymatic function regulates lipid storage and mobilization, impacting energy homeostasis and metabolic health. D-glucose can induce a seven-fold increase in ADPN mRNA levels, while insulin has a more modest effect, highlighting ADPN′s responsiveness to metabolic signals. Factors that elevate intracellular cyclic adenosine monophosphate levels can inhibit glucose-induced elevation in ADPN expression, indicating a complex regulatory mechanism. ADPN mRNA levels show inverse correlation with fasting glucose levels, suggesting higher ADPN expression associates with increased insulin sensitivity, implicating ADPN in obesity and diabetes pathophysiology. In humans, ADPN gene expression is tightly regulated by energy balance fluctuations, while in murine adipocytes, ADPN expression parallels fatty acid synthase and Srebp1c, a variant of Srebp1, underscoring ADPN′s significance in metabolic regulation.
