ADK (H-1) is a mouse monoclonal IgG2a kappa light chain antibody that detects ADK protein in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-ADK antibody (H-1) is available unconjugated or conjugated to agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® dyes. Adenosine kinase (ATP:adenosine 5-prime-phosphotransferase), or ADK, is an abundant enzyme in mammalian tissues that catalyzes gamma-phosphate transfer from ATP to adenosine, regulating extracellular adenosine and intracellular adenine nucleotide concentrations. Adenosine, an extracellular signaling molecule, influences cardiovascular, nervous, respiratory, and immune systems, with elevated levels at injury and inflammation sites. Adenosine effectively inhibits neuronal activity and suppresses seizures in various epilepsy models. The human ADK gene, located on chromosome 10q11-q24, produces two transcripts encoding 345-amino acid and 362-amino acid enzyme forms. These alternatively spliced variants differ only at the 5-prime end, where the short form′s first four residues are replaced by 21 residues in the long form. Both isoforms phosphorylate adenosine with identical kinetics and require Mg2+ for activity. ADK functions optimally under dilute conditions but forms soluble aggregates at higher concentrations, leading to enzyme inactivation.
