AChRα9 Antibody (8E4) is a mouse monoclonal IgG2a kappa light chain antibody that detects AChRα9 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-AChRα9 antibody (8E4) is available as a non-conjugated isotype antibody. AChRα9 (8E4) antibody recognizes a protein that plays a crucial role in the auditory system, particularly in cochlear hair cells, where AChRα9 is essential for mediating efferent synaptic transmission between cholinergic olivocochlear fibers and outer hair cells (OHCs). AChRα9 represents the only nicotinic acetylcholine receptor found in cochlear hair cells, with expression localized primarily in the basal regions of adult rat cochlear OHCs and in the apical regions of inner hair cells (IHCs). AChRα9 channel facilitates calcium ion influx, which is vital for the modulation of auditory signals and the protection of hair cells from damage. Understanding specific interactions and functions of AChRα9 provides insights into hearing mechanisms and potential therapeutic targets for auditory disorders.

I saw two clear bands in the WB picture showed on your website. And the isotype control is igG2a Kappa. My question is that the mAb is the full length of heavy chain and a light chain, or purified IgG2a kappa chain. Thank you.

Asked by: yu long
Thank you for your question. The Western blot data you are referring to on our website is not representative of the endogenous molecular weight for AChRα9. That is Western blot data in a human recombinant fusion protein. This is a complete IgG2a antibody. The light chain type is kappa.

For Western Blot, is it recommended to use denatured or non-denatured conditions with AChRα9 (8E4): sc-293282 antibody?

Asked by: Germaine
Thank you for your question. We recommend this antibody for use in denatured Western Blot conditions. It has not been validated for use in non-denatured conditions. Please contact our Technical Service Department for further details or inquiries.
