SEE ALSO...

AAT Antibody (H-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects AAT protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-AAT antibody (H-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AAT protein, also known as alpha-1-antitrypsin, plays a crucial role in protecting lung tissue from damage caused by neutrophil elastase, an enzyme that can lead to pulmonary emphysema when left unchecked. This 394 amino acid hepatic acute phase protein is primarily synthesized in the liver and is also expressed in macrophages, where AAT helps maintain the balance of proteolytic activity in the lungs. AAT is a highly polymorphic glycoprotein, and AAT variants exhibit distinct isoelectric-focusing patterns, which can be important for understanding individual susceptibility to lung diseases. The gene encoding AAT is located on human chromosome 14, adjacent to a related serine protease inhibitor gene that encodes corticosteroid-binding globulin. Notably, oxidation of the methionine 358 residue in AAT′s active site significantly reduces AAT′s ability to inhibit elastase, highlighting the importance of post-translational modifications in regulating function. A deficiency in AAT can lead to a 20-30 fold increased risk of developing early-onset pulmonary emphysema, making the study of AAT protein and its interactions vital for understanding respiratory health.
