

A20 Antibody (8E8.38) is a mouse monoclonal IgG1 antibody that detects A20 in human samples through applications such as western blotting (WB) and immunoprecipitation (IP). A20 is a crucial Cys2/Cys2 zinc finger protein that is upregulated in response to various inflammatory stimuli, including tumor necrosis factor (TNF) and interleukin 1 (IL-1), and serves as a negative regulator of nuclear factor kappa B (NFκB) gene expression. By inhibiting NFκB activation, A20 plays an essential role in the resolution of inflammatory responses, thereby preventing excessive cellular damage and promoting tissue homeostasis. A20′s structure is particularly significant, as A20 contains seven zinc finger domains in its C-terminal region, with only four being necessary for the in vitro inhibition of TNF-induced NFκB activation. This structural feature allows A20 to effectively interact with key signaling proteins such as TRAF2, TRAF6, and IkB kinase (IKK) γ, which are vital for modulating cell survival and apoptosis. Additionally, A20′s interaction with TXBP151, a novel A20-binding protein, suggests a mechanism through which A20 exerts anti-apoptotic effects. Given A20′s role in negative feedback regulation of signal transduction pathways, A20 and associated proteins hold promise as potential therapeutic targets for a range of inflammatory diseases.
