    5-LO Antibody (A-9) is a mouse monoclonal IgG1 antibody that detects 5-lipoxygenase (5-LO) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). 5-LO plays a crucial role in the biosynthesis of leukotrienes, which are lipid mediators that significantly influence inflammatory responses, particularly in conditions such as asthma and allergic reactions. 5-LO is primarily expressed in polymorphonuclear leukocytes, macrophages, and mast cells, where 5-LO catalyzes the initial steps in leukotriene production. Notably, 5-LO cellular localization is dynamic and varies among different cell types; for instance, in activated blood polymorphonuclear leukocytes, 5-LO translocates from the cytosol to the nuclear envelope in a calcium-dependent manner. In alveolar macrophages, the majority of 5-LO is found in the nucleus, and upon activation, 5-LO binds to the nuclear membrane, a process that is influenced by various signaling molecules and post-translational modifications, including phosphorylation. This intracellular shuttling is essential for 5-LO function, as such movement allows for precise regulation of leukotriene synthesis in response to cellular signals, highlighting 5-LO importance in mediating inflammatory processes. Anti-5-LO antibody (A-9) serves as an invaluable tool for researchers studying the mechanisms of inflammation and the role of leukotrienes in various diseases.
