    5-LO Antibody (B-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects 5-lipoxygenase (5-LO) of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-5-LO antibody (B-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. 5-lipoxygenase plays a crucial role in inflammatory response by catalyzing the first two steps in leukotriene biosynthesis, which are lipid mediators contributing to bronchoconstriction and increased vascular permeability during allergic reactions and asthma. 5-LO is predominantly expressed in polymorphonuclear leukocytes, macrophages, and mast cells, with cellular localization being particularly significant. In activated blood polymorphonuclear leukocytes, 5-LO translocates from cytosol to nuclear envelope in a calcium-dependent manner, while in alveolar macrophages, 5-LO is primarily localized in the nucleus. Upon activation, intranuclear 5-LO associates with nuclear membrane, a process influenced by various signaling molecules, phosphorylation events, and a specific nuclear localization signal encoded at the amino terminus. This dynamic intracellular shuttling of 5-LO is essential for leukotriene synthesis and highlights the importance of precise localization in regulating inflammatory responses.
