4E-BP1 Antibody (11G12C11) is a mouse monoclonal IgG1 kappa light chain antibody that detects 4E-BP1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-4E-BP1 antibody (11G12C11) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. 4E-BP1 plays a crucial role in regulating protein synthesis by acting as a translational repressor. 4E-BP1 binds to eukaryotic translation initiation factor 4E (eIF-4E), preventing formation of the eIF-4F complex essential for translation initiation. Phosphorylation of 4E-BP1 by various kinases, such as S6 kinase p70, MAP kinases, or protein kinase C, leads to dissociation from eIF-4E, promoting translation of mRNAs critical for cell growth and proliferation. This regulatory mechanism is particularly important in cancer biology, where dysregulation of protein synthesis can contribute to tumorigenesis. 4E-BP1 also interacts with 4E-BP2, which modulates eIF-4E activity, highlighting the complexity of translational control in cellular processes.

Rated 5 out of 5 by from Excellent antibody for western blottingReceived a sample and used it to probe by western blotting lysates of activated human leukocytes. Antibody worked very well. We detected two strong bands, and one faint third band. The same bands were also detected using the phospho 4E-BP1 (sc-271947) antibody.

Rated 2 out of 5 by from Works only on the unphosphorylated protein4E-BP1 protein is normally phosphorylated on several residues as part of it's regulation. at this state the antibody does not recognized the protein!!!! only after inhibiting mTOR and reducing the phosphorylation of the protein can it be recognized by the antibody

Rated 2 out of 5 by from Ok but weakThe antibody works but is very weak. Need a lot of it.

Rated 1 out of 5 by from Disappointing resultsIt took us ages to figure out why we weren't getting any signal for this antibody in western blot...finally I realized it was because it runs much higher than our other total and phospho 4E-BP1 antibodies, and we were consistently cutting off the bands. The variation, also, is much greater than for the rabbit total 4E-BP1 we have from CST, which is confusing. This variation makes me hesitant to use this antibody for any future work.
