14-3-3 γ Antibody (D-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects 14-3-3 gamma protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-14-3-3 gamma antibody (D-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. 14-3-3 gamma protein plays a crucial role in cellular signaling and regulation, particularly in cancer biology, where 14-3-3 gamma is involved in processes such as apoptosis, mitogenic signaling, and cell-cycle checkpoints. 14-3-3 gamma protein is primarily located in the cytoplasm, where 14-3-3 gamma interacts with various target proteins, facilitating proper function and localization. Cytoplasmic presence of 14-3-3 proteins is essential for sequestering and regulating ligands, which can prevent degradation and modulate enzymatic activity. The 14-3-3 family consists of seven isoforms, including 14-3-3 β, γ, ε, ζ, η, θ, and Σ, which form dimers that create binding sites for ligands sharing a common 14-3-3 consensus binding motif, often characterized by serine/threonine phosphorylation. Dysregulation or loss of 14-3-3 gamma expression can lead to various pathological conditions, underscoring 14-3-3 gamma′s importance in maintaining cellular homeostasis and signaling integrity.

Does this recognize the cleaved or uncleaved form of 14.3.3 gamma?

Thank you for your question. This mouse monoclonal antibody is specific for an epitope mapping between amino acids 128-149 within an internal region of 14-3-3 γ of human origin. We have not specifically tested the antibody to try and detect the cleaved form. Based on the cell lines that we have tested, and the Western blot data shown on our website, the antibody detects the antigen closer to the predicted molecular weight of 33 kDa.
