COL2A1 항체(M2139)는 마우스, 쥐, 인간 및 애기장대 기원의 콜라겐 II형 단백질을 WB, IP, IF 및 IHC(P)로 검출하는 IgG2b κ 마우스 단일 클론 COL2A1 항체(콜라겐 II형 알파 1 사슬 항체라고도 함)입니다. COL2A1 항체(M2139)는 비접합 항-COL2A1 항체로 제공됩니다. 광범위한 COL 유전자 제품군(콜라겐)은 피브릴 형성 간질성 콜라겐(유형 I, II, III 및 V)과 기저막 콜라겐(유형 IV)을 포함한 여러 사슬 유형으로 구성되며 각 유형은 여러 동형체를 포함합니다. 콜라겐은 인장 강도가 높은 섬유질 세포 외 기질 단백질로 힘줄과 연골과 같은 결합 조직의 주요 구성 요소입니다. 모든 콜라겐은 삼중 나선 도메인을 포함하고 있으며 복잡한 결합 조직을 형성하기 위해 종종 측면 자기 결합을 보입니다. 또한 여러 콜라겐은 정상적인 조직 구조와 기능을 유지하는 데 중요한 세포 접착에 중요한 역할을 합니다. 연골에서는 콜라겐 II형이 피브릴의 대부분을 구성합니다. 콜라겐 II형에 대한 민감화는 쥐, 생쥐 및 고등 영장류에서 류마티스 관절염 및 재발성 다발성 연골염과 유사한 침식성 다발성 관절염을 유도합니다.

I would like to detection of collagen type 2 in western blot assay. I tried dilution 1:500 in 5% skinmilk. and 2nd antibody used sc-2005 product. but It does not detection. How can I do? my sample orign is human pellet.

Thank you for the question. COL2A1 Antibody (M2139) is a mouse monoclonal mouse IgG2b κ (kappa light chain) provided at 100 µg/ml. We currently do offer Sample sizes for this product. Please request this with your supplier. However, you can also purchase the product with our 1 year full product warranty if you are planning to use this product in any of the warranted applications and warranted species: recommended for detection of Collagen Type II of mouse, rat, human and a origin by WB, IP, IF and IHC(P); also reactive with additional species, including bovine.

I would like to do some IF on my mouse samples. Do you have any suggestions on how this can be successful using a mouse monoclonal primary?

I can find the donkey anti-mouse secondary that I am accustomed to using. What do you recommend as a replacement?

Rated 1 out of 5 by from ATDC5では検出できずATDC5はマウス由来の前駆軟骨細胞株としてしられています。今回定法に従い分化誘導をかけ、回収した細胞に対してM2139を試用しました。遺伝子の発現誘導はPCRで検出されましたが、目的サイズにバンドは検出されませんでした。原因の一つとしては二次抗体が推奨のものでは無かったことが考えられます。

Rated 4 out of 5 by from It works for flow cytometry and microscopy!While the antibody has some background compared with unstained cells at 1:50 (probably helped by the facts that it is an IgG2a antibody and not an IgG1 antibody and we worked with myeloid cells), it showed staining in microscopy (with DAPI as counterstain) and a shift in intensity by flow cytometry for target-expressing cells compared to non-expressing cells! We used goat anti-mouse A488 as secondary antibody. Happy we gave this antibody a try.
