카스파제-1 항체(14F468)는 웨스턴 블롯(WB), 면역침전(IP), 면역형광 등 다양한 응용 분야에서 IL-1 전환 효소(ICE)로도 알려진 카스파제-1을 검출하기 위해 특별히 설계된 마우스 단일 클론 IgG1 카파 경쇄 항체입니다. (IF), 면역 조직 화학. caspase-1 (14F468) 항체는 인간 caspase-1의 아미노산 영역 371-390을 표적으로 삼아, 마우스, 쥐, 인간, 돼지를 포함한 여러 종에 걸쳐 전체 길이와 절단된 형태를 모두 인식할 수 있습니다. 카스파제-1은 비활성 전구체의 단백질 분해 절단을 통해 전염증성 사이토카인인 인터루킨-1β(IL-1β)와 인터루킨-18(IL-18)의 성숙을 매개함으로써 염증 반응에서 중추적인 역할을 합니다. 또한, 카스파제-1은 세포 자멸사 유도 과정에 관여하여, 프로그램된 세포 사멸 과정에서 세포의 구조적 해체에 기여합니다. 카스파제-1의 활성 형태는 효소 활성에 필수적인 p20과 p10 소단위로 구성된 4량체입니다. 인간 CASP1 유전자는 염색체 2q14에 위치하며, 간, 심장, 골격근, 신장, 고환과 같은 주요 조직에서 발현됩니다. 카스파제-1의 조절 장애는 염증, 패혈성 쇼크, 상처 치유 장애, 특정 백혈병을 포함한 다양한 병리학적 상태와 관련이 있으며, 정상적인 생리학 및 질병 상태 모두에서 그 중요성을 강조합니다. 항-카스파제-1 항체(14F468)는 카스파제-1의 신뢰할 수 있고 특이적인 검출을 제공하여 다양한 생물학적 기능과 질병 메커니즘에서의 역할에 대한 연구를 촉진합니다.

For western blots, should I dilute the primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 or just T-TBS+5%BSA? Thank you

Thank you for your question. We recommend UltraCruz® Blocking Reagent: sc-516214 for blocking purposes. In general, milk-based blocking buffers are recommended for reducing background while BSA can be used for improving blots with weak signal.

Hi, I have a question, how many band are revealed with Casp1 antibody (sc-56036)? after my treatment I saw  3 bands (45kDa; 35kDa; 25-27kda) best regards Vincenzo

Thank you for your question. Anti-caspase-1 Antibody (14F468): sc-56036 was raised against amino acids 371-390 of caspase-1 of human origin. As such it is able to recognize any of the five described isoforms for this protein. The bands you are detecting correspond to isoforms alpha, gamma, and delta.

What concentration of stimulated THP-1 cells were used for the image on the datasheet?

What are the conditions to stimulate THP-1 cells with PMA and LPS to detect CASP1 p10, similar the image of the datasheet? What is the PMA and LPS concentration? How much time?

Thank you for your question. Here are the conditions we suggest for stimulation using LPS and PMA, respectively: 5 µg per mL of LPS, induction time 2 hours. 40ng/ml of PMA, induction time 2 hours.
