What dilution do you recommend starting with for flow cytometry using the fluorophore-conjugated version of this antibody? Are there any publications using this antibody for flow cytometry?

sc-166940这只c-fos的抗体，我染小鼠脑组织的冰冻切片，0.3%Triton打孔10分钟，山羊血清室温封闭一小时，一抗4℃过夜，使用Alexa Fluo 488的二抗室温避光孵育1小时，但无阳性信号出现，请问问题出在哪里了呢？

What temperature is suitable for storing and reusing c-fos antibody (Anti-Fos/c-Fos Antibody (E-8) X) following dilution, 4C, or -20C freezer?

I have a question about Anti-Fos/c-Fos Antibody (E-8) X (catalog: sc-166940 X) for western blot analysis. I would like to know if it is possible to use both HRP and fluorescent dye conjugated secondary antibodies for detecting the sc-166940 X antibody.

Thank you for your question. This primary antibody can be detected by either fluorescence-, or by an HRP-based secondary detection. Our secondary binding proteins m-IgG1 BP-HRP: sc-525408 and m-IgG1 BP-CFL 488: sc-533661 have been optimised for detecting primary mouse IgG1 antibodies including this one.

Hello, The c-Fos Antibody (K-25): sc-253 was discontinued and replaced by c-Fos Antibody (E-8): sc-166940? In what way are they the same? sc-253 has been shown to work for fish c-fos detection, so will sc-166940 work in the same way? Cheers!

Thank you for your question. c-Fos Antibody (E-8): sc-166940 is a mouse monoclonal IgG1 antibody that was raised against the same peptide antigen that was used to develop the rabbit polyclonal antibody, c-Fos Antibody (K-25): sc-253. The mouse monoclonal antibody has also been published in zebrafish samples, and is recommended to work for zebrafish. For some examples of literature using this mouse monoclonal antibody in zebrafish, please refer on our website to the product citations with PMID's: 30222997, 29626809 and 29061647.

The conjugated primary antibody is recommended and warrantied for flow cytometry (FCM), however we are still gathering data to show on our website. Check back for updates!

I would like to determine the c-Fos expression in the mouse whole brain cleared with CUBIC or iDISCO. Have you tried to use E-8 for that?

I had used the same protocol for Egr1 ChIP (a similar inducible transcription factor) and it works nicely. Also, quite some background in WB.
