Spot 14 Antibody (F-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects Spot 14 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Spot 14 (F-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Spot 14, also known as S14 or thyroid hormone responsive protein (THRSP), is a small acidic protein primarily located in the nucleus, where Spot 14 plays a crucial role in regulating lipid metabolism. Spot 14 is predominantly expressed in tissues involved in triglyceride synthesis, such as liver, mammary glands, and adipose tissues. Spot 14 (F-7) monoclonal antibody is essential for studying growth and differentiation, as Spot 14 functions as a transcriptional regulator for genes encoding proteins involved in lipogenesis. Spot 14 expression is activated by various lipogenic stimuli, including thyroid hormone, dietary carbohydrates, insulin, and glucose, while being downregulated by catecholamines and glucagon. Spot 14 expression exhibits oscillation in accordance with circadian clock, and Spot 14 knockdown impairs lipid synthesis and induces apoptosis. In many breast cancers, Spot 14 is overexpressed, suggesting a role in promoting cell growth and survival, making Spot 14 a significant target for research in cancer biology and metabolic disorders.

Dear Santa Cruz, my question is relative to the HepG2 cells stained with Spot 14 Antibody (F-7): sc-137178. Is it possible to know if the cells have been pre-treated with tween or similar and Ab incubation time and concentration? Thanks a lot.

Asked by: zoe80
Thank you for your question. You can find the protocol used for this image here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining We recommend a starting dilution of 1:50 for IF for this antibody, with a 1 hour incubation. If you have any further questions, please contact Technical Service.

What is the recommended fixation method for immunofluorescence staining with Spot 14 (F-7): sc-137178 monoclonal antibody?

Asked by: TinTin
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
