Iba1 Antibody (F-4) is a mouse monoclonal IgG3 antibody that detects ionized calcium-binding adapter molecule 1 (Iba1) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Iba1 is a crucial 147 amino acid cytoplasmic protein that binds calcium and plays a significant role in macrophage activation and function, making Iba1 essential for immune responses and tissue repair. Iba1 contains two EF-hand domains, which are vital for calcium-binding capability, and is induced by various cytokines and interferons, highlighting Iba1′s importance in inflammatory processes. In unstimulated state, Iba1 colocalizes with actin, but upon activation, Iba1 translocates to lamellipodia, facilitating cellular movement and response to injury. Additionally, Iba1 serves as a specific marker for human microglia and is expressed by macrophages in injured skeletal muscle, underscoring Iba1′s relevance in neuroinflammation and muscle regeneration. The gene encoding Iba1 is located on chromosome 6p21.33 within the tumor necrosis factor gene cluster, which is part of the human major histocompatibility complex, further emphasizing Iba1′s role in immune regulation and potential implications in various diseases.
