L'anticorpo anti-istone H3 (1G1) è un anticorpo monoclonale IgG di topo che rileva l'istone H3 in campioni di topo, ratto e uomo mediante western blotting (WB) e immunofluorescenza (IF). L'anticorpo anti-Istone H3 (1G1) è disponibile in forma non coniugata. L'istone H3 svolge un ruolo cruciale nell'organizzazione strutturale della cromatina, essenziale per il confezionamento del DNA all'interno del nucleo. L'istone H3 costituisce una parte centrale della struttura ottamerica del nucleosoma che avvolge il DNA, regolando l'espressione genica e la replicazione del DNA. L'istone H3 mantiene la stabilità genomica e facilita i processi di trascrizione e riparazione. L'istone H3 subisce diverse modifiche post-traslazionali, tra cui la metilazione e l'acetilazione, che influenzano le interazioni tra DNA e proteine, incidendo sulla struttura e sulla funzione della cromatina. Queste modifiche regolano l'espressione genica e hanno un impatto sulla differenziazione cellulare e sulla risposta ambientale. Le capacità di rilevamento dell'anticorpo anti-istone H3 (1G1) rendono questo reagente prezioso per lo studio delle dinamiche della cromatina e della regolazione epigenetica in vari contesti biologici.

I performed ICC with this antibody in mitotic cells, but I couldn't detect histone H3 signal co-localized with the DAPI signal. Could you help me solve this issue?

Just received a free sample, which dilution would you recommend for nuclear extracts WB?

Thank you for your question. Dilution recommendations can be found in the product's datasheet, which is available right here on the product page. For western blots we recommend a starting dilution of 1:200, and a dilution range between 1:100-1:1000. When working with nuclear extracts, an amount of protein between 10–20 µg is appropriate.

Rated 5 di 5 di da H3 Ab for Western BlottingAsked for a sample before purchasing. Worked straight away on CRC cell lines HCT116 and DLD1 (drug treated and untreated) when loading 20 ug of whole protein lysates and using 1:500 dilution in 5% BSA/TBST. Would definitely recommend.

Too strong signal with the dilution you recommend. Conditions need to be better set. Apparent MW: about 15kDa.
