Prolyl-tRNA synthetase (ProRS), also known as PARS, is an enzyme that in humans is encoded by the EPRS gene. This enzyme belongs to the family of aminoacyl-tRNA synthetases (aaRSs), which play a critical role in protein synthesis.Aminoacyl-tRNA synthetases are responsible for charging tRNA molecules with their corresponding amino acids, a crucial step in the process of translation. ProRS specifically attaches the amino acid proline to its corresponding tRNA molecule (tRNA^Pro). This prolyl-tRNA then participates in the ribosome-mediated synthesis of a protein, by incorporating proline into the growing polypeptide chain at the appropriate positions as determined by the mRNA template.
In humans, ProRS is part of a bifunctional enzyme complex that also contains glutamyl-prolyl-tRNA synthetase. This bifunctional enzyme is unique among aaRSs and is linked by a sequence that is homologous to a region of the vesicular transport protein SecY.Beyond their classical role in protein synthesis, aminoacyl-tRNA synthetases, including ProRS, have been found to have additional functions in cellular processes such as cell signaling, RNA splicing, and the regulation of apoptosis.
