

GILZ Antibody (D-2) is a mouse monoclonal IgG1 antibody that detects glucocorticoid-induced leucine zipper (GILZ) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). GILZ plays a crucial role in mediating the immunosuppressive effects of glucocorticoid hormones, particularly in T cells, where GILZ expression is induced by dexamethasone. GILZ protects T cells from apoptosis induced by anti-CD3 antibodies by inhibiting the expression of Fas and Fas ligand, thereby promoting cell survival. GILZ′s ability to interact with key transcription factors, including Egr-2, Egr-3, and NFAT/AP-1, allows GILZ to modulate gene expression critical for T cell function. Notably, GILZ binds to c-Fos and c-Jun, which inhibits the binding of active AP-1 to its DNA consensus site, thus affecting downstream signaling pathways. Additionally, GILZ′s interaction with NFkB subunits prevents their nuclear translocation, further influencing immune responses. The gene encoding human GILZ is located on chromosome Xq22.3, and GILZ expression is predominantly found in normal lymphocytes from the thymus, spleen, and lymph nodes, while being absent in nonlymphoid tissues such as the brain, liver, and kidney.
