Tuberin Antibody (B-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects tuberin protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-tuberin antibody (B-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Tuberin, also known as Tuberous sclerosis complex (TSC) subunit 2, plays a crucial role in cellular signaling pathways, particularly in regulating cell growth and proliferation. Tuberin (B-5) monoclonal antibody serves as a key component of the TSC complex, which acts as a tumor suppressor by inhibiting the mammalian target of rapamycin (mTOR) pathway. Dysregulation of this pathway due to mutations in the TSC2 gene, which encodes tuberin, can lead to the development of hamartomas and other tumors associated with Tuberous sclerosis complex, a genetic disorder characterized by various benign tumors and neurological issues. The interaction between tuberin and hamartin is essential for maintaining cellular homeostasis, and disruptions in this interaction can result in significant pathological consequences. Anti-tuberin antibody (B-5) serves as an invaluable tool for researchers studying TSC and related signaling pathways, providing insights into the molecular mechanisms underlying tumorigenesis and potential therapeutic targets.

What is the total protein amount should we use to detect by tuberin Antibody (B-5): sc-271314?

Asked by: THUY ANH
Thank you for your question. We recommend using 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane on an SDS gel.

Can tuberin (B-5): sc-271314 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Asked by: jerojero
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, tuberin (B-5): sc-271314 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
