Synaptotagmin V Activators

Synaptotagmin V (SytV) is a synaptic vesicle protein crucial for neurotransmitter release at neuronal synapses. SytV functions as a calcium sensor, playing a pivotal role in calcium-dependent exocytosis. Upon calcium influx into the presynaptic terminal, SytV undergoes conformational changes due to calcium binding to its tandem C2 domains. These conformational changes enable SytV to interact with the SNARE complex and the synaptic vesicle membrane, facilitating membrane fusion and neurotransmitter release into the synaptic cleft. Additionally, SytV has been implicated in regulating synaptic vesicle trafficking and recycling, highlighting its multifaceted role in neurotransmission. Activation of Synaptotagmin V primarily involves the elevation of intracellular calcium levels within the presynaptic terminal. Calcium influx into the terminal occurs through voltage-gated calcium channels in response to action potentials, or through calcium release from intracellular stores such as the endoplasmic reticulum. The increased calcium concentration leads to the binding of calcium ions to the C2 domains of SytV, triggering its activation and subsequent facilitation of synaptic vesicle fusion. Furthermore, activation of SytV may be modulated by protein-protein interactions with other synaptic proteins, phosphorylation events, or post-translational modifications, although the precise details of these regulatory mechanisms require further elucidation.
