SPLUNC2 Activators

SPLUNC2 (Short Palate, Lung, and Nasal Epithelium Clone 2), also known as BPIFA2 (Bactericidal/Permeability-Increasing Fold-Containing Family A Member 2), is a protein that plays a crucial role in the maintenance of human airway epithelium. Abundantly expressed in the airway epithelium, SPLUNC2 is believed to contribute significantly to the host defense mechanism and hydration. It possesses antimicrobial properties and is involved in the regulation of surfactant homeostasis. Although the precise mechanisms of regulation and function of SPLUNC2 are not completely understood, they are a subject of active research across the scientific community.

A variety of chemical compounds are thought to potentially stimulate the expression of SPLUNC2. These include 1,25-Dihydroxyvitamin D3, an active form of Vitamin D, and Retinoic Acid, a metabolite of Vitamin A, both of which play a role in immunomodulatory responses and epithelial cell differentiation respectively. Lithium Chloride is another compound that, through its stimulation of numerous cellular processes, could lead to the enhanced expression of SPLUNC2. Dexamethasone, a corticosteroid, may encourage the expression of SPLUNC2 as part of its orchestration of inflammatory and immune responses. Other compounds such as Curcumin, Resveratrol, Epigallocatechin Gallate, Quercetin, Sulforaphane, Genistein, Berberine, and Naringenin, all possess antioxidant properties and could induce an increase in SPLUNC2 expression. However, the exact mechanisms through which these compounds stimulate SPLUNC2 expression are not fully understood and may be indirect.
