SLURP1 Activators

Secreted LY6/PLAUR domain-containing 1 (SLURP1) is a protein that is part of the Ly6/uPAR family characterized by the presence of the LU domain, which is a structural motif found in several cell-surface receptors and other secreted proteins. SLURP1 was identified as a secreted protein that is expressed in various human tissues, including the skin, and has been implicated in the regulation of immune responses and cellular signaling.The LY6/uPAR domain is known for its role in various biological processes, including cell adhesion, movement, and in the case of uPAR (urokinase plasminogen activator receptor), in the regulation of proteolysis and cell-surface plasminogen activation. Members of this family are typically attached to the cell membrane through a glycosylphosphatidylinositol (GPI) anchor, but SLURP1, as suggested by its name, is secreted.

SLURP1 has been particularly associated with the modulation of nicotinic acetylcholine receptors (nAChRs), which are ligand-gated ion channels that mediate fast signal transmission at synapses. Through its interaction with nAChRs, SLURP1 can influence keratinocyte proliferation and differentiation, suggesting a role in the maintenance of skin homeostasis and integrity.Mutations in the gene encoding SLURP1 have been linked to Mal de Meleda, a rare autosomal recessive skin disorder characterized by diffuse palmoplantar keratoderma, hyperkeratosis, and an increased risk of squamous cell carcinoma of the skin.This condition often present with transgressive keratoderma, lichenoid skin lesions, and various nail abnormalities.
