S-100A3 Activators encompass a diverse range of chemical compounds that indirectly promote the functional activity of S-100A3 through the modulation of calcium signaling and interaction with other cofactors. Zinc sulfate and magnesium chloride contribute to this process by serving as essential cofactors that stabilize the S-100A3 protein and ensure proper functioning of enzymes in pathways where S-100A3 is active.

The elevation of intracellular calcium levels by calcimycin and ionomycin directly enhances the calcium-binding capability of S-100A3, thereby potentially amplifying its role in calcium-dependent signaling. Similarly, PMA and forskolin act upstream to modify the phosphorylation landscape, which can have downstream effects on the activity and interactions of S-100A3.
