RSP3 Activators are a set of compounds that primarily function through the modulation of the cAMP signaling pathway. Compounds such as Forskolin, 8-Br-cAMP, and Isoproterenol function by increasing the intracellular levels of cAMP, either through direct activation of adenylate cyclase or by acting as an agonist at the beta-adrenergic receptor. Elevated cAMP levels lead to the activation of protein kinase A (PKA), which phosphorylates RSP3, leading to its functional activation. H-89 dihydrochloride, on the other hand, works by inhibiting PKA, increasing the pool of unphosphorylated PKA that can then phosphorylate and activate RSP3.
Other compounds within this class, such as Rolipram, IBMX, Vinpocetine, Zaprinast, Cilostazol, Pentoxyfylline, Theophylline, and Papaverine, function by inhibiting various types of phosphodiesterases, which are enzymes responsible for the breakdown of cAMP. By inhibiting these enzymes, these compounds effectively increase the intracellular levels of cAMP. As with the previous group of compounds, this increase in cAMP leads to the activation of PKA, which then phosphorylates and activates RSP3.
