RNF184 Inhibitors

RNF184, an E3 ubiquitin-protein ligase, partakes in the complex regulation of protein ubiquitination and degradation. The inhibitors presented influence several junctures in the protein degradation process and, while not direct inhibitors of RNF184, they provide a glimpse into how RNF184's function can be modulated. For instance, MG132 and Lactacystin, as proteasome inhibitors, obstruct the final stages of the ubiquitin-proteasome pathway. By doing so, they can influence the overall ubiquitin landscape and the fate of proteins tagged by E3 ligases such as RNF184. Further upstream, PYR-41 targets the E1 ubiquitin-activating enzyme, and its inhibition can echo downstream to RNF184's function by impeding the initiation of the ubiquitination cascade.

MLN4924, which inhibits the NEDD8-activating enzyme, presents an interesting angle as it hampers cullin-RING ligases, a subclass of E3 ligases. It highlights the broad spectrum of ubiquitination and its intricate regulation. Another intriguing facet of protein regulation is autophagy, a catabolic process where cells devour their own components. Compounds like Chloroquine and 3-Methyladenine, by disrupting autophagy, can cast an indirect influence on the fate of ubiquitinated proteins and the broader impact of E3 ligases like RNF184.
