PRAS40 Activators

PRAS40 activators are a class of chemical compounds that exert their influence on the PRAS40 (Proline-rich Akt substrate of 40 kDa) protein by modulating its activity through various signaling pathways. One prominent group of PRAS40 activators includes compounds that target the mTORC1 (mammalian target of rapamycin complex 1) pathway. Rapamycin, for instance, is a well-known PRAS40 activator that can inhibit mTORC1, relieving PRAS40 from suppression and ultimately leading to its activation. Similarly, amino acids such as leucine can activate mTORC1, which, in turn, phosphorylates and inactivates PRAS40. These compounds function as key regulators of PRAS40 activity by modulating mTORC1-dependent signaling cascades.

Additionally, other PRAS40 activators indirectly impact PRAS40 through the PI3K-AKT pathway. Insulin can activate the PI3K-AKT pathway, which subsequently phosphorylates and inactivates PRAS40. Chemicals like Wortmannin and LY294002, both PI3K inhibitors, can disrupt this pathway, indirectly leading to PRAS40 activation. Furthermore, PRAS40 can be influenced through AMPK activation by compounds such as 2-Deoxy-D-glucose (2-DG) and AMP (Adenosine Monophosphate), resulting in mTORC1 inhibition and the consequent release of PRAS40 from its inhibitory role.
