PKR Activators

PKR Activators constitute a varied group of chemicals that specifically target and activate Protein Kinase R (PKR), a serine/threonine kinase that is usually triggered by double-stranded RNA (dsRNA). PKR activation leads to the phosphorylation of the eukaryotic translation initiation factor 2 alpha (eIF2α), which ultimately inhibits protein synthesis and cell growth. These activators range from synthetic analogs of dsRNA, such as Polyinosinic-polycytidylic acid (Poly I:C), to small molecules like 2-Aminopurine, to even natural compounds like Resveratrol and Curcumin. While the natural molecules often come from dietary sources such as fruits and spices, synthetic variants are typically designed to mimic stressors or triggers that naturally induce PKR activation. Additionally, compounds originally intended for other biological targets, such as certain anticancer agents like Cisplatin and Docetaxel, have also been found to induce PKR activation.The mechanisms through which these activators engage PKR are diverse, mirroring the complexity of the pathways in which PKR itself is involved. Some chemicals activate PKR by directly binding to its kinase domain, while others may work indirectly, activating cellular pathways that, in turn, lead to the activation of PKR. For instance, Imiquimod works via toll-like receptors to induce PKR activation, whereas Anisomycin causes cellular stress that leads to PKR activation. The chemical structures of PKR activators are equally varied, ranging from complex organic molecules to simpler purine and pyrimidine analogs. Despite this variety, what unites them is their ability to initiate a cascade of cellular events via PKR activation, affecting various cellular functions like protein synthesis, cell growth, and the stress response.
