Pim-3 Inhibitors

Pim-3 inhibitors are a class of compounds that might reduce the activity of Pim-3 kinase through various mechanisms, including direct inhibition of kinase activity, interference with signaling pathways that regulate Pim-3 expression, and prevention of protein translation. For example, non-specific kinase inhibitors such as quercetin and staurosporine could potentially inhibit Pim-3 activity by binding to its active site. Specific inhibitors like SMI-4a and AZD1208 are designed to target the kinase activity of Pim family members and could directly inhibit Pim-3. PI3K and mTOR inhibitors, such as LY294002 and rapamycin, might decrease Pim-3 expression by modulating the signaling pathways that control cell growth and survival, which often regulate the expression of Pim kinases. In addition to these, inhibitors that affect protein translation, such as silvestrol, could reduce Pim-3 protein levels, thereby decreasing its activity in the cell. Compounds like gossypol could bind to the ATP-binding pocket of Pim-3, preventing ATP from binding and thereby inhibiting its kinase activity.
