Osteopontin (OPN) is a multifunctional phosphorylated glycoprotein that plays diverse roles in various physiological and pathological processes. Expressed in a variety of tissues and cell types, OPN participates in cell adhesion, migration, and signaling. Its interaction with specific integrin receptors and CD44 variants on the surface of cells facilitates these functions. Due to its involvement in processes such as wound healing, inflammation, immune responses, and bone remodeling, OPN has been studied extensively in cell biology and biochemistry. In particular, its roles in cellular adhesion and migration have made it a protein of interest in studies of tissue remodeling and tumor metastasis.

OPN inhibitors are chemical entities designed to target and modulate the activity or expression of OPN. Given that OPN can act as a ligand for certain cell surface receptors, these inhibitors might function by preventing the binding of OPN to its receptors, thus interfering with downstream signaling and cellular responses. Alternatively, some OPN inhibitors might act at the transcriptional or translational level, reducing the overall expression of OPN in the cell. Another mechanism of action for these inhibitors is to interfere with post-translational modifications of OPN, such as phosphorylation, which can influence its activity and interactions with other proteins.
