Chemical activators of Olr185 can be understood through their distinct mechanisms of influencing intracellular signaling pathways that lead to the protein's activation. Calcium Ionophore A23187 directly increases intracellular calcium concentration, a critical second messenger, which can induce a conformational change in Olr185, activating the protein. Similarly, BAY K8644 activates L-type calcium channels, causing a surge in intracellular calcium that can activate Olr185. Thapsigargin disrupts calcium sequestration by inhibiting the sarco/endoplasmic reticulum Ca2+-ATPase (SERCA), consequently raising cytosolic calcium levels that activate Olr185. Ionomycin also facilitates the increase of intracellular calcium by selectively transporting calcium ions into cells, activating Olr185 through calcium-dependent pathways.
Further activation of Olr185 can occur through modulation of kinase activity. Forskolin activates adenylate cyclase, resulting in elevated cAMP levels that activate protein kinase A (PKA).
