NAT-15 Activators

NAT-15 activators comprise a diverse set of chemical compounds that indirectly enhance the functional activity of NAT-15 through various signaling pathways. Forskolin and 8-Bromo-cAMP both increase cellular cAMP levels, leading to the activation of PKA which may phosphorylate NAT-15 or its associated regulatory proteins, resulting in heightened activity. Similarly, PMA as a PKC activator, and Rolipram as a PDE4 inhibitor, also elevate the phosphorylation status within the cell which could indirectly result in the enhanced activity of NAT-15. Ionomycin and A23187, both calcium ionophores, raise intracellular calcium levels and can potentially activate calcium-dependent kinases that modify the activity or stability of NAT-15. Spermine, while primarily known for its role in modulating ion channels, may affect NAT-15 by altering its local cellular environment, thereby influencing its functional state.

The activity of NAT-15 is further influenced by the modulation of various kinase signals. EGCG, as a kinase inhibitor, can relax the phosphorylation-based inhibition of NAT-15, leading to its enhanced activity. LY294002, by inhibiting PI3K, and SB216763, by inhibiting GSK-3, bothalter downstream signaling in a way that could indirectly facilitate an increase in NAT-15 activity through changes in the phosphorylation landscape.
