Med9 Inhibitors

Inhibitors of Med9 like Triptolide which is a diterpenoid epoxide that can disrupt the assembly or function of the Mediator complex, thereby affecting Med9 function. Flavopiridol and BI-1347 are inhibitors of CDK9, which is part of P-TEFb, a critical factor for transcription elongation that the Mediator complex regulates. I-BET151, JQ1, BRD4 Inhibitor JQ1, SGC-CBP30, and GSK2801 are inhibitors of BET bromodomain proteins, which recognize acetylated lysine residues on histone tails and are important for the regulation of gene expression, potentially influencing the function of Med9 by altering the chromatin context in which the Mediator complex operates. Cortistatin A and THZ1 target CDK8 and CDK7, respectively, both of which are kinases associated with the Mediator complex and are important for the regulation of transcription by phosphorylating components of the transcription machinery, including the C-terminal domain of RNA polymerase II. CX-4945, a CK2 inhibitor, affects this kinase that can phosphorylate Mediator complex subunits, including Med9. Pladienolide B, by affecting the spliceosome, has the potential to alter RNA processing and indirectly the transcriptional landscape that Med9 is involved in.
