MC-CPA Inhibitors

Chemical inhibitors of MC-CPA play a crucial role in the direct inhibition of this serine protease by binding to its active site and preventing substrate interaction. Benzamidine, for instance, serves as a competitive inhibitor, occupying the active site of MC-CPA, which results in the prevention of substrate cleavage, a similar mechanism of inhibition is exhibited by Soybean trypsin inhibitor (SBTI) and Aprotinin, both of which form complexes with serine proteases, leading to reversible inhibition. Phenylmethanesulfonyl fluoride (PMSF) and Diisopropylfluorophosphate (DFP) take a more permanent approach by covalently modifying and thus irreversibly inactivating the serine residue at the active site of MC-CPA. This alteration halts the enzymatic activity of MC-CPA by rendering it incapable of hydrolyzing peptide bonds in its substrates.

Further expanding the arsenal of MC-CPA inhibitors, chemicals like 4-(2-Aminoethyl) benzenesulfonyl fluoride hydrochloride (AEBSF), Gabexate mesylate, Nafamostat mesylate, and Camostat mesylate interact with the active site of MC-CPA. AEBSF performs irreversible inhibition by binding to the active serine residue, while Gabexate mesylate, Nafamostat mesylate, and Camostat mesylate inhibit by blocking the access of substrates to the active site of MC-CPA. α1-Antitrypsin functions by a similar mechanism, where it attaches to the active site, preventing substrate cleavage. On the other hand, Leupeptin and Chymostatin inhibit MC-CPA by reversibly binding to the active site, which impedes the protease's catalytic function.
