KIF25 Inhibitors

Chemical inhibitors of kinesin family member 25 (KIF25) encompass a range of small molecules that can impede its function by targeting related motor proteins involved in spindle dynamics and microtubule interactions. Monastrol, a small molecule inhibitor, can disrupt the activity of kinesin Eg5, a protein related to KIF25, by specifically inhibiting its motor activity which is crucial for mitotic spindle function. This disruption can affect KIF25 indirectly, as it may alter the microtubule dynamics upon which KIF25 operates. Similarly, S-Trityl-L-cysteine targets the ATPase activity of kinesin Eg5, potentially affecting the cellular processes that KIF25 is involved in by modifying the microtubule network. Another inhibitor, K858, disrupts the interaction of mitotic kinesin Eg5 with microtubules, which can compromise the microtubule architecture and, consequently, the function of KIF25 in cell division. CW069, targeting the allosteric inhibition of kinesin-5, can disrupt bipolar spindle formation, which is critical to KIF25's role in cellular processes. Additionally, GSK923295, an inhibitor of centromere-associated protein-E (CENP-E), can alter mitotic progression, which is a process where KIF25 is also known to participate, thereby affecting its function. All these inhibitors, through their targeted action on related proteins, can influence the microtubule network and spindle apparatus that are essential for the normal operation of KIF25.
