Similarly, phorbol 12,13-dibutyrate (PDBu) function as a synthetic analog of diacylglycerol (DAG), also directly stimulating PKC, which in turn can lead to the mobilization of granzyme A. Forskolin, by elevating cAMP levels through the activation of adenylate cyclase, can influence signaling pathways that promote the release of granzyme A.
Additionally, ionophores like ionomycin, A23187 (calimycin), and thapsigargin can alter intracellular calcium concentrations, which are crucial second messengers in the activation process of cytotoxic cells, thereby facilitating the release of granzyme A. Calcium ionophores increase the intracellular calcium levels, which can lead to the activation of downstream kinases and phosphatases that ultimately result in the degranulation of cytotoxic cells. Thapsigargin, a SERCA pump inhibitor, causes a sustained increase in cytosolic calcium levels, further enhancing this process. Moreover, prostratin and ingenol 3,20-dibenzoate, similar to PMA, activate PKC and thus can contribute to the cellular mechanisms that lead to the secretion of granzyme A. BIM (Bisindolylmaleimide I), although primarily a selective inhibitor of PKC, at certain concentrations can activate PKC, which could influence the activation and degranulation of cytotoxic lymphocytes to secrete granzyme A.
