GPR80 Activators

GPR80, also known as OXGR1, is predominantly activated by α-ketoglutarate (α-KG). This metabolite is a pivotal component in various metabolic pathways, particularly the tricarboxylic acid (TCA) cycle. As a result, numerous metabolites and compounds that feed into or modulate the TCA cycle can influence the levels of α-KG, and thus, indirectly activate GPR80. Such chemicals include glutamate, isocitrate, fumarate, malate, and citrate, which are either precursors or derivatives of α-KG within the cycle. As the TCA cycle's progression directly impacts α-KG levels, agents like NADH and acetyl-CoA, which feed into the cycle, can indirectly promote GPR80 activation.

Apart from the direct influence of α-KG, the cell-permeable variant, dimethyl α-ketoglutarate, can increase intracellular α-KG levels, leading to GPR80 activation. Other chemicals like AMP and adenosine can potentially modulate metabolic pathways, influencing α-KG concentrations. Specifically, their role in monitoring cellular energy states could adjust the TCA cycle flux, thus adjusting α-KG levels. In summary, while α-KG is a direct activator of GPR80, a spectrum of metabolic intermediates and modulators, particularly those linked to the TCA cycle, can influence the activation of GPR80 indirectly.
