Get4 Activators

Get4, also known as Golgi to ER Traffic Protein 4, is a protein involved in the process of protein targeting to the endoplasmic reticulum (ER). It's a component of the GET (Golgi-to-ER trafficking) complex, which is responsible for the delivery of tail-anchored (TA) proteins to the ER in both yeast and mammalian cells.TA proteins have a single transmembrane domain near their C-terminus and play crucial roles in various cellular processes, including vesicular trafficking, cell signaling, and protein translocation. However, the hydrophobicity of the transmembrane domain makes the delivery and insertion of TA proteins into the ER membrane a challenging process, which is where the GET complex comes in.

Get4 plays a central role within this complex. It forms a subcomplex with Get5 (also known as Ubl4A in mammals) and binds to Sgt2 (small glutamine-rich tetratricopeptide repeat-containing protein), which initially recognizes and binds the TA proteins. Get4 is essential for the handover of TA substrates from Sgt2 to the main GET complex for their subsequent insertion into the ER.In summary, Get4 is a crucial component of the GET complex, playing a key role in the delivery of tail-anchored proteins to the endoplasmic reticulum. Its function is critical for maintaining cellular homeostasis and proper functioning, given the wide range of cellular processes that TA proteins are involved in.
