FANK1 Inhibitors

Chemical inhibitors of FANK1 can achieve functional inhibition through various mechanisms by targeting signaling pathways that FANK1 is known to be part of. Staurosporine, a broad-spectrum protein kinase inhibitor, can hinder FANK1's kinase activity by preventing the phosphorylation of its substrates, which is a crucial step for its function. Similarly, 5-Iodotubercidin, as an adenosine kinase inhibitor, can block the phosphorylation of adenosine, potentially disrupting FANK1. LY294002 and Wortmannin, both PI3K inhibitors, can suppress the PI3K/Akt signaling pathway. PD98059 and U0126, which target MEK within the MAP kinase pathway, can prevent the activation of FANK1.

Moreover, SP600125 acts as a JNK inhibitor and can functionally inhibit FANK1. SB203580, by inhibiting p38 MAP kinase, can also interfere with FANK1 activity. Rapamycin, an inhibitor of mTOR, can suppress FANK1 function. PP2, which inhibits Src family kinases, would disrupt Src kinase signaling, subsequently affecting FANK1 activity. Lastly, Gefitinib, which specifically inhibits the EGFR tyrosine kinase, would impede FANK1 activity.
