eIF4AI Activators

The chemicals listed above are not direct activators but may influence eIF4AI activity indirectly through their impact on the translation initiation process or the general machinery of protein synthesis. eIF4AI functions as an essential component of the eIF4F complex, facilitating the unwinding of mRNA secondary structures during the initiation phase of protein synthesis. Compounds like Hippuristanol, Silvestrol, Rocaglamide, and Pateamine A specifically target eIF4A, the family to which eIF4AI belongs. By inhibiting eIF4A's interaction with mRNA, these compounds can indirectly affect the function of eIF4AI in translation initiation.

Other chemicals such as Emetine, Cycloheximide, Puromycin, Homoharringtonine, Anisomycin, and 5-Fluorouracil are known to inhibit various stages of protein synthesis. While these do not target eIF4AI directly, their impact on the overall process of protein synthesis can influence the functional context in which eIF4AI operates. Additionally, Actinomycin D, which inhibits RNA synthesis, and Rapamycin, an inhibitor of the mTOR pathway that regulates protein synthesis, can also indirectly affect eIF4AI activity. These agents alter the cellular environment and the availability of substrates and components necessary for translation initiation, thereby potentially impacting eIF4AI function.
