DCAF15 Activators represent a group of chemical compounds that play a pivotal role in enhancing the functional activity of DCAF15 within the cell. These activators operate through various distinct mechanisms, each contributing to the heightened functionality of DCAF15. For instance, Nordihydroguaiaretic Acid (NDGA) stabilizes DCAF15 and promotes its interaction with the E3 ubiquitin ligase complex. This stabilization prevents DCAF15's degradation, resulting in an increased presence of DCAF15 in the complex. Consequently, this leads to improved substrate recognition and ubiquitination, ultimately enhancing its role in protein regulation. In a similar vein, Resveratrol indirectly activates DCAF15 by altering the cellular redox environment. This modulation enhances DCAF15's activity, particularly its participation in the Keap1/Nrf2 pathway, crucial for cellular responses to oxidative stress.

Coenzyme Q10, another activator, acts as a co-factor for DCAF15, facilitating its binding to target proteins. This co-factor role enhances DCAF15's ability to regulate protein stability and degradation, effectively increasing its functional activity within the cell. Staurosporine, on the other hand, activates DCAF15 through the activation of specific kinase pathways. It phosphorylates DCAF15, leading to conformational changes that enhance its interaction with target substrates, thereby promoting their degradation.
