Cyclophilin 40 Inhibitors

Cyclophilin 40 (Cyp40) is a member of the immunophilin family, proteins that bind to immunosuppressive drugs such as cyclosporin A. However, beyond its interaction with immunosuppressants, Cyp40 plays a crucial role in protein folding and trafficking, serving as a co-chaperone with heat shock proteins (HSPs), particularly HSP90. This interaction is vital for the proper folding and function of a wide array of protein substrates, many of which are involved in signal transduction pathways, including steroid hormone receptors and kinases. Cyp40 possesses peptidyl-prolyl cis-trans isomerase (PPIase) activity, enabling it to catalyze the isomerization of peptide bonds at proline residues, a reaction critical for the proper folding of proteins. Through these mechanisms, Cyp40 is implicated in numerous cellular processes, including cell signaling, protein homeostasis, and the response to stress, underscoring its significance in maintaining cellular function and integrity.

The inhibition of Cyp40 involves blocking its PPIase activity or disrupting its interaction with partner proteins such as HSP90. This can be achieved through small molecules that bind directly to the active site of Cyp40, disrupting it from engaging in its enzymatic function of prolyl isomerization. Inhibiting Cyp40 can lead to a cascade of effects within the cell, primarily due to the accumulation of misfolded proteins and the disruption of cellular signaling pathways that rely on the proper function of Cyp40-associated proteins. Such inhibition is of particular interest in the study of diseases characterized by protein misfolding and aggregation, as well as in cancer, where the chaperone function of Cyp40 is exploited by rapidly dividing cells to maintain protein homeostasis.
