Spastin Antibody (A-2) is a mouse monoclonal IgG2a light chain antibody that detects Spastin in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Spastin is a member of the AAA protein family, characterized by its ATPase domain, and plays a crucial role in various cellular processes, including intracellular motility, membrane trafficking, and organelle biogenesis. This 616 amino acid protein is predominantly localized in the nucleus and cytoplasm, where Spastin is essential for microtubule dynamics, influencing the stability and organization of the cytoskeleton. Proper functioning of Spastin is vital, as mutations in the Spastin gene (SPAST, SPG4) are linked to the most common form of hereditary spastic paraplegia, specifically spastic paraplegia 4. This autosomal dominant disorder is characterized by progressive weakness and stiffness in the lower extremities due to retrograde axonopathy affecting corticospinal motor neurons. Notably, SPAST-specific mutations account for approximately 40% of all autosomal dominant hereditary spastic paraplegias, underscoring Spastin′s importance in maintaining neuronal health and function. Anti-Spastin antibody (A-2) serves as an invaluable tool for researchers studying the molecular mechanisms underlying these neurological disorders and the broader implications of Spastin in cellular dynamics.
