Rap1GAP Antibody (G-1) is a mouse monoclonal IgG3 antibody that detects Rap1GAP in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Rap1GAP plays a crucial role in cellular signaling by specifically stimulating the GTP hydrolytic activity of the monomeric G protein Rap1, which is essential for regulating various cellular processes, including cell proliferation and differentiation. Rap1GAP is predominantly located in the brain, kidney, and pancreas, where Rap1GAP acts as a signal integrator, coordinating Gz signaling and Rap1 signaling pathways. Notably, a novel isoform of Rap1GAP, known as Rap1GAPII, has been identified to bind specifically to Gαz, a member of the Gi family of trimeric G proteins. This interaction blocks the ability of regulators of G protein signaling to stimulate GTP hydrolysis of the α subunit, thereby modulating downstream effects on adenylyl cyclase activity. Furthermore, stimulation of the Gi-coupled M2 muscarinic receptor leads to Rap1GAPII translocation from cytosol to membrane, resulting in a decrease in GTP-bound Rap1 and subsequent activation of the ERK/MAPK signaling pathway. anti-Rap1GAP antibody (G-1) is an invaluable tool for researchers studying these complex signaling mechanisms and their implications in various physiological and pathological contexts.
