PDK2 Antibody (180CT10.2.3) is a mouse monoclonal IgG1 antibody that detects pyruvate dehydrogenase kinase 2 (PDK2) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. PDK2 plays a crucial role in cellular metabolism by regulating the activity of the pyruvate dehydrogenase complex (PDC), which is essential for converting pyruvate into acetyl-CoA, a key substrate for the citric acid cycle. The regulation of PDC by PDK2 determines whether cells utilize glucose or fatty acids for energy production, particularly under conditions of nutrient deprivation. PDK2 is predominantly expressed in heart and skeletal muscle tissues, where energy demands are high, and PDK2′s activity is modulated by various metabolic signals, including the availability of nutrients. PDK2′s structure features five conserved regions that are critical for kinase activity, and PDK2 shares mechanistic similarities with bacterial His-kinases, highlighting evolutionary significance. PDK2′s ability to phosphorylate the E1α subunit of PDC leads to the inactivation of the complex, thereby influencing metabolic pathways and energy homeostasis in response to physiological changes.
