Hemoglobin γ Antibody (B-4) is a mouse monoclonal IgG3 antibody specific for an epitope mapping between amino acids 9-43 near the N-terminus of human Hemoglobin gamma. Hemoglobin gamma, also known as fetal gamma globin or HBG1/HBG2, is a crucial component of fetal hemoglobin (Hb F), which consists of two alpha (α) chains and two gamma (γ) chains. During fetal development, Hemoglobin gamma interacts with alpha globin chains to form Hb F tetramers, facilitating efficient oxygen transport from the mother to the fetus due to its higher affinity for oxygen compared to adult hemoglobin. Proteins such as BCL11A and KLF1 interact indirectly with Hemoglobin gamma by regulating its gene expression. BCL11A acts as a repressor that silences gamma globin expression after birth, leading to the switch from fetal to adult hemoglobin. KLF1, a transcription factor, also plays a role in this switch by activating BCL11A expression, thereby contributing to the repression of gamma globin genes. Understanding these interactions is important for developing therapeutic strategies for hemoglobinopathies like sickle cell disease and beta-thalassemia, where reactivation of gamma globin expression can ameliorate disease symptoms. Hemoglobin γ (B-4) antibody is validated for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) applications, making it a valuable tool for studying the role and regulation of Hemoglobin gamma in human biology.
