GAD-65抗体(A-3)是一种IgG1 κ型小鼠单克隆GAD-65抗体(也称为GAD65抗体或GAD2抗体),可通过WB、IP、IF、IHC(P)和ELISA检测鼠源、大鼠源和人源的GAD-65蛋白。GAD-65抗体(A-3)既可以是非偶联的抗GAD-65抗体形式,也可以是多种偶联形式的抗GAD-65抗体,包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®偶联物。在大脑中发现两种形式的谷氨酸脱羧酶(GADs):GAD-65(也称为GAD2)和GAD-67(也称为GAD1、GAD或SCP)。GAD-65和GAD-67是II型脱羧酶家族蛋白的成员,负责催化从L-谷氨酸产生GABA(γ-氨基丁酸)的限速步骤。尽管两种GADs都存在于大脑中,但GAD-65定位于神经末梢的突触囊泡膜上,而GAD-67则分布于整个细胞中。GAD-67负责GABA合成的基础水平。在神经传递中对GABA需求增加的情况下,GAD-65会短暂激活以协助GABA的产生。GAD-65的丧失是有害的,可能会损害GABA的神经传递,然而GAD-67的丧失则是致命的。由于选择性剪接,GAD-67存在两种异构体,即主要的GAD-67形式和次要的GAD-25形式。GAD-25在大脑中不表达,但可以在各种内分泌组织中找到。

Can GAD-65 (A-3): sc-377145 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, GAD-65 (A-3): sc-377145 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 1 out of 5 by from Poor antibodyTested the antibody in immunofluorescent staining on human iPSC-derived neurons at 1:50 dilution. The antibody seems to give aspecific staining. It stains some synapses and all cell bodies of the culture, which is king of strange because only around 10-15% of these cells are expected to be GABAergic neurons. Image shows sc-377145 Mouse anti-GAD65 (green), rabbit anti-Synapsin 1 (red), chicken anti-TubB3 (blue), nuclei are counterstained with DAPI (gray).
