dystrophin抗体(H-5)是一种IgG2b κ小鼠单克隆dystrophin抗体,可通过WB、IP、IF、IHC(P)和ELISA检测小鼠、大鼠和人类来源的dystrophin。dystrophin抗体(H-5)可以是非偶联抗dystrophin抗体形式,也可以是多种偶联形式的抗dystrophin抗体,包括琼脂糖、辣根过氧化物酶、藻红蛋白、异硫氰酸荧光素和多种Alexa Fluor®偶联物。Dystrophin-glycoprotein复合物(DGC)通过细胞膜界面将肌纤维内表面的F-Actin细胞骨架连接到周围的细胞外基质。这种蛋白的缺乏会导致Duchenne(DMD)和Becker(BMD)肌营养不良。人类dystrophin基因长度为2.4兆碱基,具有80多个外显子,产生14 kb的mRNA,并包含至少八个独立的组织特异性启动子和两个poly A位点。dystrophin mRNA可以经历差异性剪接,并产生一系列编码大量蛋白质的转录本。Dystrophin占肌张力蛋白总量的约0.002%,定位于骨骼肌的三角联接处,被认为可以影响钙离子的家庭稳态和力传递。
