

CD223 (C9B7W) is a rat monoclonal IgG1 antibody. CD223 (Lymphocyte activation gene-3, LAG-3) functions as a high affinity MHC class II ligand present on CD4(+)CD8(+) T cells and NK cells. CD223 shares structural homology with CD4 molecules, featuring four similar extracellular Ig-like domains and structural motifs between D1-D3 and D2-D4 domains. A glutamic acid-proline (EP) repetitive sequence found in CD223 appears in other functionally distinct mammalian, parasitic, and bacterial proteins, suggesting influence on a conserved biological function. CD223(+)CD4(+)CD8(+) T cells associate with T cell receptor (TCR) and downregulate TCR signaling in vitro. CD223 regulates CD4-dependent T cell function through its cytoplasmic domain. A conserved "KIEELE" motif containing Lys-468 in CD223 plays an essential role in interactions with downstream signaling molecules.
