caspase-2 Antibody (1C10) is a mouse monoclonal IgG2b antibody that detects caspase-2 in human samples through applications such as western blotting (WB) and immunoprecipitation (IP). Caspase-2 is an aspartate-specific cysteine protease that plays a critical role in the apoptotic process, acting as a key regulator of programmed cell death. The unique structure of caspase-2, characterized by a long prodomain and DEXD substrate specificity, allows caspase-2 to function both as an upstream initiator and a downstream executor of apoptosis, making caspase-2 essential for maintaining cellular homeostasis. Notably, caspase-2 is predominantly expressed in the brain during development, with lower levels found in adult tissues, and localizes to various cellular compartments, including the mitochondria, Golgi apparatus, cytoplasm, and nucleus. The presence of two isoforms, caspase-2L and caspase-2S, produced through alternative splicing, further highlights this complexity; while caspase-2L promotes apoptosis, caspase-2S serves as a negative regulator, illustrating the delicate balance of apoptotic signaling pathways. Following apoptotic stimuli, the caspase-2L precursor is cleaved to generate active fragments, including p30, p18, and p12, which are crucial for executing the apoptotic program. Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
