PDC-E2 Antibody (B-2) is a mouse monoclonal IgG2a kappa light chain antibody that detects PDC-E2 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). PDC-E2 (B-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PDC-E2 plays a crucial role in the pyruvate dehydrogenase multienzyme complex, which is essential for converting pyruvate into acetyl-CoA, a key metabolic intermediate that fuels the citric acid cycle. This function is vital for energy production and metabolic regulation in cells, particularly in tissues with high energy demands such as the heart and brain. The presence of autoantibodies against PDC-E2 is a hallmark of primary biliary cirrhosis, a chronic autoimmune liver disease characterized by the destruction of bile ducts, leading to cholestasis and liver damage. PDC-E2 features a unique structure with both an amino-terminal lipoyl-bearing domain and a carboxy-terminal catalytic domain, which are critical for enzymatic activity and interaction with other proteins in the metabolic pathway. The immunodominant autoepitopes of the autoantigens correspond to the inner lipoyl domain, highlighting PDC-E2′s importance in both normal physiology and autoimmune pathology.

I am using PDC-E2 (B-2): sc-271534 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
