O anticorpo N-Ras (F155) é um anticorpo monoclonal N-Ras de camundongo IgG1 (também designado anticorpo NRAS) que detecta a proteína N-Ras de origem de camundongo, rato e humana por WB, IP, IF e IHC(P). O anticorpo N-Ras (F155) está disponível como anticorpo anti-N-Ras não conjugado. A família de genes Ras de mamíferos (também designada v-Ha-Ras, Harvey rat sarcoma viral oncogene homolog, HRAS1, K-Ras, N-Ras, RASH1 ou c-bas/has) consiste nos genes Harvey e Kirsten Ras (c-H-Ras1 e c-K-Ras2), um pseudogene inativo de cada um (c-H-Ras2 e c-K-Ras1) e o gene N-Ras. Os três oncogenes Ras, H-Ras, K-Ras e N-Ras, codificam proteínas com ligação GTP/GDP e atividade GTPase. As proteínas Ras alternam entre uma forma inativa ligada ao GDP e uma forma ativa ligada ao GTP, ativada por um fator de troca de nucleotídeos de guanina (GEF) e inativada por uma proteína ativadora de GTPase (GAP). A nomenclatura Ras tem origem na caracterização de sequências de DNA humano homólogas a fragmentos de DNA clonados contendo sequências oncogênicas de um retrovírus de mamífero tipo C, a cepa Harvey do vírus do sarcoma murino (HaMSV), derivado do rato. Em condições normais, os membros da família Ras influenciam o crescimento celular e os eventos de diferenciação em um sistema de sinalização baseado na compartimentalização da membrana subcelular. A Ras oncogênica pode desregular os processos que controlam a proliferação e a apoptose das células. A superfamília Ras de proteínas de transmissão de sinal acopladas à hidrólise de GTP pode ser subclassificada nas famílias Ras, Rho, Rab e ARF.

Hi, can this N-Ras antibody(F155) be used to detect the Nras encoded by mutated NRAS, such as NrasQ61R?

Thank you for your question. This antibody is raised against recombinant N-Ras p21 of human origin, recommended for detection of N-Ras p21 of mouse, rat and human origin by WB, IP, IF and IHC(P), we didn't test it on mutated NRAS.

Hello, I am interested in "N-Ras Antibody (F155): sc-31" for Western blot use. What were the duration, buffers, Ab concentrations, and temperature/conditions (for blocking/probing) used to QC this Ab? Thank you in advance

Thank you for your question. If you are using ECL for detection, we recommend blocking (PVDF membrane) in 5% nonfat milk + TBST for 1 hour at room temperature. We recommend using the same buffer (freshly prepared) as a diluent for both primary and secondary antibodies. If you are using Near-Infrared (NIR) Detection methods, then we recommend using our UltraCruz Blocking Reagent: sc-516214 instead of milk or BSA to reduce background. Recommended starting dilution for Western blot for the primary antibody is 1:200 (range 1:100-1:1,000). We recommend incubating primary antibody for 1 hour at room temperature, or overnight at 4C. Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue?

No. Since no direct conjugates are available for this antibody, we would not recommend using it for IHC in mouse tissue. See pan Ras (C-4): sc-166691 for N-Ras antibody conjugates, including AC, HRP, FITC, PE, Alexa Fluor® 488 and Alexa Fluor® 647.

Rated 4 out of 5 by from Application to ICCI have tested this Ab for the immunocytochemical detection of NRAS overexpressed in a thyroid-derived cell line. SIgnals were detected from cells transfected with the NRAS expression plasmid, although their intensity was low. No signals were observed in the non-transfected cell lines; therefore, detection of endogenous NRAS may be difficult.
